How We Can Reverse Aging

Reverse aging is an appealing notion to those seeking eternal youth, yet no single strategy or practice can turn back time completely. But consistent habits may support cellular health, metabolic flexibility and physical resilience over time.

Cellular senescence

Cellular senescence is a state in which permanent cell cycle arrest is initiated by DNA damage accumulation. It differs from quiescent cells which can reenter the cell cycle, and terminally differentiated ones which no longer proliferate, in that senescent cells typically exhibit a “Senescence-Associated Secretory Phenotype”, producing proinflammatory cytokines, chemokines, and growth factors to promote tissue repair and regeneration.

SASP is an endogenous and exogenous stress response mechanism, including oncogenic signals, telomere shortening, and persistent DNA damage. Senescent cells also undergo extensive chromatin reorganization in response to growth arrest; their cells typically display bright DAPI staining as well as immunoreactivity with H3K9Me2/3 histone H3 di- or trimethylated histone H3 (H3K9Me2/3) immunoreactivity; this pattern corresponds with silencing genes associated with proliferation such as E2F and cyclin A.

Persistent DNA damage triggers either the DNA repair machinery, apoptosis or both; but depending on its physiological context and extent of damage, can also induce cellular senescence. For example, radiation and chemicals that activate ATM and ATR pathways cause this form of cellular senescence while genetic disorders resulting in early aging (Werner syndrome and Hutchinson-Gilford progeria) also increase DNA damage and cause premature cellular aging through increased DNA damage and senescence.

Senescent cells can be distinguished from dividing cells by their large size, flattened shape and the presence of senescence-associated chromatin foci (SAHF). Furthermore, increased lysosomal content and disrupted nuclear envelope integrity are hallmarks of senescent cells; multinucleated and with dysfunctional mitochondria are other indicators. Senescent cells also often display signs of lactic acid fermentation phenotype, as evidenced by elevated activity of enzyme b-galactosidase at pH 6.0.

Senescent cells can be identified through a transgenic mouse model expressing luciferase, red fluorescent protein, and herpes simplex virus thymidine kinase under the minimal p16 promoter to allow selective elimination. By eliminating senescent cells selectively, lifespan was extended without impacting normal cell proliferative capacity or signs of aging being noticed; stem cell-derived exosomes containing miR-302b injection was shown to reverse cellular senescence thus prolonging lifespan while increasing physical performance and cognition.

Inflammation

Inflammation is part of our bodies’ natural response to infections, injuries or illnesses; it’s the reason that an ankle sprain looks red, swells up and feels warm when touched. Additionally, inflammation helps trap germs or toxins so the body can begin healing itself; but prolonged or frequent inflammation can become dangerous as we age; chronic inflammation has been linked to heart disease, cancer, Alzheimer’s and rheumatoid arthritis among other health problems.

Your immune system responds to injuries or infections by sending out inflammatory cells and cytokines to trap germs or toxins, starting the healing process, and beginning healing itself. This phase is known as inflammation. Your body responds in this way when something that shouldn’t be there such as bacteria in a cut wound or virus attacks occur, forcing an inflammatory response.

As we age, our levels of inflammatory molecules increase, leading to chronic inflammation as well as more senescent cells which further aggravate it and ultimately aging us further. Scientists have identified various strategies to decrease chronic inflammation such as avoiding foods which increase your risk for inflammation as well as eating plenty of plant-based food, lean proteins, whole grains and healthy fats to support optimal health.

Skin aging

There are various strategies available to prevent skin aging. These include limiting exposure to the sun and smoking as well as following an effective skin care regime with regular hydration and gentle treatments. While such diseases are part of the natural aging process, they can be prevented and treated through healthy lifestyle choices – for instance by getting regular exercise, eating nutritiously balanced diet and prioritizing sleep – which reduces your risk.

Some individuals appear to experience age-related illnesses more rapidly due to having a faster biological age (the rate at which their cells are degenerating). Any treatment to reverse aging would need to reduce this biological age in order to treat age-related diseases before they arise.

One potential solution may involve using plasma from young donors, which contains growth factors known to restore cells back to a state closer to youth. Unfortunately, however, this approach has yet to be shown effective for human consumption; further study will likely be required before becoming available on the market.

Other researchers are conducting studies aimed at reversing aging effects through gene modification. While not as advanced as Yamanaka’s approach, gene therapy shows promise for reversing age-related diseases in future. Scientists have already used gene therapy to rejuvenate old mice and extend their lifespan and prepare human clinical trials with this technology. We can all look forward to a world in which injuries and diseases are more effectively treated while our dream of whole-body rejuvenation becomes a reality and we begin living longer, healthier lives!
